I am in Kansas City right now conducting my semi-annual frog and reptile workshop. This is an Amazon milk frog I captured with a 50mm macro lens plus one extension tube to fill much of the frame with the small amphibian. The lighting comes from a ring flash, and throughout the entire workshop I use f/32 for maximum depth of field. I set the camera on manual exposure mode, the flash is on ETTL which provides the correct exposure, the shutter was set to 1/125, and the ISO was 500. I used manual focus, and I fine tuned the focusing by moving closer to and farther from the frog. Due to the extreme magnification, it's tough to decide exactly where to focus. If I focused on the eyes, the protruding mouth would be out of the plane of focus. That's why the smallest lens aperture is necessary assuming complete depth of field is the goal. With small subjects that offer beautiful and intriguing detail, you don't want to compromise on the DOF.
